DNA methyltransferases in gastric carcinogenesis


Gastric cancer is still the third most common cause of cancer-related death worldwide. While inflammation is a well-established driver of gastric tumorigenesis, only a small subset of gastric cancer patients responds to checkpoint immunotherapy. One mechanism of immune escape is silencing tumor antigen expression. DNA methyltransferases (DNMTs) are the enzymes responsible for epigenetic silencing of gene expression and are often overexpressed in tumors. One part of my project focuses on the role of DNMT3A in gastric cancer mouse models and its potential as a therapy target. In addition, I am investigating if non-responding gastric tumors can be sensitized to immune checkpoint inhibitors by the treatment with epigenetic inhibitors
